- - In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test with rats (OECD TG422, GLP), no effects were observed in estrous cycle, copulation index, fertility index, gestation length, and delivery status, but four out of 10 animals died during a gestation period in the high dose group (300 mg/kg), there were a decrease in delivery index and a decreased number of corpus lutea, accompanied by tendencies of low numbers of implantations and total offspring. On observation of lactation, other than total litter loss in one maternal animal found in the 300 mg/kg group, a low number of live offspring due to deaths of maternal animals and low viability index on day 4 due to the total litter loss were observed (Safety Test (Ministry of Economy, Trade and Industry) (Access on Sep. 2012)). The adverse reproductive effects above were not used for the rationale for classification because they were findings only at the high dose where high mortality was seen in maternal animals. In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test with rats (OECD TG422, GLP), 3/12 males and 7/12 females died in the 300 mg/kg group (converted guidance value: about 150 mg/kg/day), and they showed principal symptoms such as salivation, irregular respiration, a decrease in locomotor activity, tonic convulsion, and clonic convulsion. Pathological examination revealed an increase in hemosiderin deposition in the spleen, hemosiderin deposition of Kupffer cells in the liver, and myocardial degeneration in the dead animals and a maternal animal that had a total litter loss in the 300 mg/kg group, and an increase in hemosiderin deposition in the spleen was also observed in males in the 60 mg/kg group (converted guidance value: about 30 mg/kg/day). On hematological examination, decreases in erythrocyte counts, hemoglobin levels, and hematocrit values and increases in platelet counts and reticulocyte counts were observed. From the above, because an increase in hemosiderin deposition in the spleen and hemosiderin deposition of Kupffer cells in the liver at 300 mg/kg/day suggested the possibility of hemolytic anemia, and such a tendency was also observed in males at 60 mg/kg/day (equivalent to the guidance values for Category 2), it was classified in Category 2 (blood system). Besides, signs such as convulsions were only found in the 300 mg / kg group, which exceeded the guidance value for Category 2, and myocardial degeneration seen in the dead animals in the 300 mg/kg group was not observed in the other animals.
