Positive results were reported in the chromosome aberration test using bone marrow of rats by the inhalation exposure (in vivo mutagenicity test in somatic cells) (SIDS (2006)). Positive results were also indicated in the dominant lethal test using rats by oral or inhaled administration for 2.5 months (in vivo mutagenicity test in germ cells) (SIDS (2006)), but information on the testing method and detail of results were unknown. From the results mentioned above, effects of this substance on the somatic cells were clear, however, findings of effects on germ cells were insufficient. Therefore, the substance was classified as Category 2 based on the positive results of the chromosome aberration test of rats (in vivo mutagenicity test in somatic cells) described above. As relevant information, negative results were reported in the micronucleus test using rats by 2-week inhalation exposure (in vivo mutagenicity test in somatic cells) (OECD TG474, GLP-compliance) (SIDS (2006)). As for in vitro studies, positive results were reported in the Ames test and the gene mutation test using CHO cells (SIDS (2006), NTP DB (Access on Jan. 2011)). - - In the developmental toxicity study in rats by inhalation exposure during the organogenetic period, no adverse effects were observed in reproductive index and development of the fetus in the high dose group that maternal body weight gain was significantly reduced. Thus, this material was judged to show neither the embryotoxic nor teratogenic potential (SIDS (2006)). On the other hand, it was reported that in the oral or inhalation studies in male rats for 2.5 months, in addition to abnormalities and necrosis in the cells of the spermatogenic epithelium were observed, increased preimplantation mortality of embryos after mating of treated males with untreated females (SIDS (2006)). However, it was reported that detail of the test method and result were not mentioned and the documentation was insufficient for assessment (SIDS (2006)). Therefore, since data on sexual functions and fertility were insufficient, and no data on exposure for females were available, the substance was classified as "Classification not possible". As relevant information, in another study, an increase in the incidence of postimplantation death was reported for rats that inhaled during the pregnancy, however, this data was concluded as insufficient for assessment (SIDS (2006)). In addition to occurrence of deaths, toxic symptoms containing spasm, inappetence and weakness were observed in the acute oral toxicity study in rats at the dose levels of 119 and 297 mg/kg. Moreover, clinical symptoms consisting of spasm with gasping and flaccid paralysis were observed in the acute vapor inhalation toxicity study in rats at the concentration of 47 mg/L (SIDS (2006)). Since the exposure amounts in either study were equivalent to Category 1 of the guidance values, the substance was classified as Category 1 (nervous system). Further, in the acute inhalation (as vapors) study in rats, occurrence of deaths, irregular respiration, lacrimation and salivation were observed at the concentrations 0.364 mg/L and above, and lesions and hemorrhages in the lungs and trachitis were noted in histopathological examinations (SIDS (2006)). In another acute inhalation (as vapors) study in rats, marked reduction in respiratory rate was observed at 0. 15 mg/L and above, and the damage of epithelium of bronchus was found at 2.13 mg/L/30 min (0.74 mg/L/4hrs) and above, respectively (SIDS (2006)). All of the effective doses corresponded to Category 1 of the guidance values, thus the substance was classified as Category 1 (respiratory system). Furthermore, in the inhalation studies described above, cell degenerations in the liver, spleen, thymus and lymph nodes, or renal damage were observed concurrently (SIDS (2006)). Based on the information, since the effects by administration of the substance might extend systemically to the multiple organs, the substance was classified as Category 1 (systemic toxicity). Additionally, it was reported, in the human case, that depression of central nervous might occur by the exposure via oral and inhalation at low concentration, and coma might occur rapidly in inhalation at high concentrations (HSDB (2006)). In the 4-week inhalation study in rats exposed by vapors, concentration dependent effects on respiratory epithelium were observed, and inflammations and focal ulcerations were noted in mucous membranes of the nasal to respiratory tract and bronchi in the treatment groups at 8 and 12 ppm/6hrs (converted dose level as that of 90-day study: 0.014 and 0.021 mg/L/6hrs) (SIDS (2006), ACGIH (2001)). In another rat inhalation study exposed by vapors for up to 19 months, hyperplasia of basal cell in the nasal cavity and focal atrophy in the mucous membranes of the nasal cavities were observed in the group at 1 ppm/6hrs (0.0052 mg/L/6hrs) after 3 months of exposure (SIDS (2006)). As mentioned above, since the dose levels were all within Category 1 of the guidance values, the substance was classified as Category 1 (respiratory organ system). Besides, the effects on the liver were reported in the 26-week oral study in rats (RTECS (2009)). However, this information was not adopted as the evidence to classify due to information from the "List 3" information source designated in the GHS classification guidance for the Japanese government with its details unspecified.
