In the rabbit test (OECD TG 404), 4-hour application of 500 mg of the substance as paste form with saline solution to 3 rabbits caused mild erythema and edema in one animal, moderate erythema and edema in the second one, and severe erythema and edema in the third one 1 hour to 3 days after patch removal. All signs of irritation were reversible 7 days after removal of the patch, and authors evaluated the substance as slightly irritating (BUA Report 225 (2000)). The substance was classified as Category 2 based on the document that two of three animals exhibited more than moderate irritation 1 hour to 3 days after patch removal. In the rabbit eye irritation test (OECD TG 405), instillation of 100 mg of the substance to conjunctival sac caused severe reddening and swelling of conjunctiva and corneal opacity 1 to 7 hours after application. The disappearance of these effects were very slow but were reversible after 21 days, and authors evaluated the substance as irritating to eyes (BUA Report 225 (2000)). In humans, there were many case reports of occurrence of allergic contact dermatitis and positive results in the patch tests with both patient and healthy volunteers (BUA Report 225 (2000), HSDB (2006)). In the guinea pig maximization test, 60 - 80% of treated animals showed positive response after the initial challenge. At rechallenge, 80% of the animals showed positive reaction, and the authors concluded that the substance had strong sensitizing potential (BUA Report 225 (2000)). This substance is used as preservative in cutting oil, paints, adhesives and for leather. A contact dermatitis may be caused in hairdressers and shoes manufacturers, and the substance was listed as a skin sensitizer (Contact Dermatitis, Frosch, 4th (2006)). In the study that 50 mg/kg of the substance was subcutaneously administered to the rats of 2 strains (CD and BDIX) on days 13 or 14 of gestation, no developmental effects of prenatal fetus were observed, postnatal deaths were observed in about half of the pups in both strains (BUA Report 225 (2000)). In addition, the EU classification for the substance was "GHS Repro cat. 2". Therefore, the substance was classified as Category 2. As relevant information, in the study that 20 mg/kg of the substance was intraperitoneally administered to the rats on day 7 or 11 and 12 of gestation, no effects on the fetus were observed (BUA Report 225 (2000)). In addition, the substance was prohibited from using in Canada in 2009, the evaluation was necessary when placing it back on the market. - - In two 3-month feeding studies in rats, significant reduction in testicular weight and dysfunction of spermatogenesis was reported at the highest dose level of 50 mg/kg/day (BUA Report 225 (2000)). However, testicular effect was treated as adverse in the classification for reproductive toxicity, therefore it was not object of this hazard class. As for the other effects, there were no reports describing adverse effects. The effects at the upper limit of the guidance values (100 mg/kg/day) or higher were unknown. Furthermore, in the 30-day dermal application study in rabbits, fatty infiltration of the liver and heart, and deposition of hemosiderin in the spleen were reported at the dose level of 100 mg/kg/day (converted dose level as that of 90-day study: 33.3 mg/kg/day) or higher (BUA Report 225 (2000)). However, since no effects were seen in hematology and no details for the degree and toxicological significance of the histopathological changes were known, "Classification not possible" was concluded.
