There is a description that in a skin irritation test with rabbits, when this substance was applied for 2 hours, mild irritation was seen at a primary skin irritation index of 1.87 (SIDS (2009)). In addition, it was described that in two tests in which this substance was applied to rabbits for 24 hours, it was moderately irritating at a primary skin irritation index of 4.16, and it was slightly irritating at a primary skin irritation index of 1.83 (both ECETOC JACC (1996), SIDS (2009)). There are descriptions that in an eye irritation test (OECD TG 405 compliant) with rabbits, one animal exhibited slight conjunctival redness, edema and discharge, but the redness resolved after 24 hours as did other irritation findings after 48 hours, and the remaining animals showed no irritation. Also, it is described that there was no primary irritation index (SIDS (2009)). In addition, there are reports that in other multiple eye irritation tests with rabbits, slight irritation was observed (ECETOC JACC (1996), SIDS (2009)). There is no single-exposure information on this substance in humans. As for experimental animals, in single inhalation exposure tests with rats and mice with this substance or n-butyl methacrylate (CAS RN 97-88-1), there is no detailed description of the dose where symptoms were observed, but there is a report that as a symptom common to both substances, lung noise, irregular respiration, and nasal discharge were observed, and it is described that it is thought to be due to exposure to substances irritant to the respiratory tract (ECETOC JACC (1996)). In the oral route, there is a report that in a single oral dose test with rats, hypoactivity, staggering gait, ataxia, decreased muscle tone of extremities and abdomen, diarrhea, piloerection, and decreased body temperature were observed (SIDS Dossier (2009)). However, it was not adopted as evidence of the classification since the minimal dose of the test was 8,880 mg/kg, above the Category 2 and there is no information on the dose below that. Therefore, this substance was classified in Category 3 (respiratory tract irritation). Besides, IUCLID (2000) used for the previous classification could not be confirmed because it was not available.
