It is reported that in a skin irritation test using rabbits, moderate edema and moderate to severe necrosis were observed after this substance (170 to 250 mg/kg) was locally applied, and moderate chemical burns were found after application of a 10% aqueous solution (ACGIH (7th, 2001)). Moreover, it is reported that moderate irritation was found after open application of an isomer mixture containing 75% this substance (25% 1-hydroxy-2-propyl acrylate) to rabbit skin (DFGOT vol. 16 (2001)). Furthermore, it is reported that severe deep-seated necrosis was observed after 15-minute or 4-hour occlusive application of an isomer mixture containing 66% this substance (33% 1-hydroxy-2-propyl acrylate, 1% free acrylic acid or 1-hydroxy-3-propyl acrylate) (animal species: unknown) (DFGOT vol. 16 (2001)). As information for an isomer mixture of propyl acrylate (this substance = 74%, 1-hydroxy-2-propyl acrylate = 25%, acrylic acid = 0.73%), in a 1-month (20 to 21 administration) inhalation toxicity test using dogs, rats, rabbits, and mice, effects on the respiratory system such as squamous epithelial metaplasia and ulceration in the mucous membranes of the nasal concha, mucopurulent rhinitis, nasal mucosa inflammation, laryngitis, tracheitis, bronchitis, and pneumonia were observed at 5 ppm (converted to a Guidance value equivalent: 0.006 mg/m3) within a range of Category 1 or higher in dogs, rats, and rabbits (DFGOT vol. 16 (2001)).
