aranciamycin生合成gene cluster(ara-cluster)のクローニングとシークエンス報告。

ORF21(427aa): GT(AraGT)

nucleotide-activated deoxysugarsを産生可能なStreptomyces fradiae A0, Streptomyces diastatochromogenes Tu6028でara clusterを発現。

その結果、C-8でmethoxy基を欠いているが、C-7で2-O-methyl-L-rhamnose, L-rhodinose, L-axenose, D-amicetoseをそれぞれ含む新しい化合物aranciamycin A, B, C, Dが検出された。

araGTを不活化したときはglycosylateされたaranciamycin派生体を産生しなかった。

よって、AraGTは上記の糖残基すべての転移を担うことが示され、その基質特異性がゆるいことがわかる。

MaTu and MCF7 cells増殖に対して、L-rhodinose、D-amicetoseを含むaranciamycin B, Dは最高の抑制活性を示した。
