mtmLはPKSの上流領域に含まれる。
CoA誘導体として有機酸や長鎖FAを活性化するacyl CoA ligasesに似ている。

acetyl-CoAは芳香族polyketidesの前駆体であり、ふつうは一次代謝によって生成されるが、mithramycin生産株はmithramycin経路にacetateを向かわせる効率的なシステムを持ち、そこでMtmLはacetate loading enzymeとして使用されるかもしれない。
