[Rationale for the Classification] As in vivo test results on this substance, there are no positive reports in a dominant lethal test, but results showing chromosomal aberrations in germ cells (2) and results showing somatic cell mutagenicity (3) were obtained. From the above, because there are results indicating germ cell mutagenicity in part of animal tests, it was classified in Category 1B. [Evidence Data] (1) In a dominant lethal test on this substance, there are no results by which it is clearly judged to be positive (ATSDR (2012), IARC 58 (1993), DFGOT Vol. 22 (2006)) (2) It is reported that after administrating this substance, chromosomal aberrations in mouse spermatocytes and hamster oocytes and altered gene expression in mouse spermatid occurred (ATSDR (2012), IARC 58 (1993), DFGOT Vol. 22 (2006)). (3) It is reported that as the result of administrating this substance to mice, dose-dependent increases in the incidences of micronuclei, sister chromatid exchanges, and chromosomal aberrations were observed in the peripheral blood. Moreover, it is reported that increased single strand breaks in the DNA were observed in rats after administrating cadmium chloride (EU-RAR (2007), DFGOT Vol. 22 (2006), NICNAS IMAP (Accessed Dec. 2018)). [Reference Data, etc.] (4) It is reported that increases in the incidences of micronuclei and sister chromatid exchanges in peripheral lymphocytes were shown in workers occupationally exposed to cadmium (compound not specified) and that human lymphocytes from people inhabiting cadmium-polluted areas of China were found to have increased micronuclei rates and a higher frequency of chromosomal aberrations and severe aberration types. However, conflicting knowledge is also reported (ATSDR (2012)). (5) As for in vitro, positive results are reported in a chromosomal aberration test and a sister chromatid exchange test in mouse spleen cells and a DNA single-strand break assay in Escherichia coli (NICNAS IMAP (Accessed Jan. 2019)). (6) Water solubility of this substance is 1,400 g/L (20 deg C) (ATSDR (2012), EHC 135 (1993), DFGOT Vol. 22 (2006)), and it is very soluble in water. (7) This substance was classified in Muta. 1B in EU CLP. |
