rifamycin PKSのmodule 6のAT domainを rapamycin PKSのmodule 2のATで置換したA. mediterranei S699 mutants(rifAT6::rapAT2)は、24-desmethylrifamycin B と 24-desmethylrifamycin SVを産生した。

こうして得られた24-desmethylrifamycin Bを変換して得た24-desmethylrifampicinは、rifampicin耐性M. tuberculosis株に対して優秀な抗菌活性を示した。

Competition feeding experimentsによりrapamycin PKSの本当のstarter unitとして化合物4(4,5-dihydroxycyclohex-1-enecarboxylic acid)を同定。

NRPSとの類似性から、化合物4はPKSのLMにあるCL(carboxylic acid ligase) domainによってAMP活性化され、LMのACPに移され、それから隣接するER domainによって還元され、鎖伸長を開始するKS1 domainへ移動されると提唱されている。

化合物4が還元されたDHCHCも直接取り込まれたので、CL domainの基質特異性は広い。
