rapQONMLのdeletion mutantはpipecolateを添加すると、rapamycinの代わりにanalog 16-O-desmethyl-27-desmethoxyrapamycinを産生した。これは、rapNによってコードされるP450 hydroxylaseがC-27に特異的で、rapQとrapM によってコードされるO-methyltransferasesがC-16 and C-27でのhydroxyl基をメチル化することを示す。

rapKIJMNOQL deletion mutant S.hygroscopicus MG2-10をrapJNOQLで相補して、starterとしてcyclohexanecarboxylic acidを与えると、C-27にmethoxy基のある化合物を産生。
rapKが単独でS.lividans TK24に発現されても、C-27 OH基が形成されないので、pre-rapamycin(未修飾aglycon)は修飾されなかった。
他の遺伝子組み合わせでの相補実験結果もふまえ、RapQはC-27 OH基に特異的なO-methylationを担うと言える。
