Fredericamycin(FDM) gene clusterの同定論文。

fdmW: holo-ACP synthase

FdmWは、resistomycin生合成gene clusterのputative ACPS-type PPTase=RemDにとても似ている。
apo-ACP FdmH の転写後修飾のためのPPTasesをたぶんコードする。

fdmC-fdmT3のfragmentをS.albusへ導入し、FDM-A産生に十分であることを確認している。

fdmWの不活化はFDM産生を約93%減少、相補で回復。
fdm clusterのACP(fdmH)と他のACPsとの反応との比較から、FdmWがFDM生合成に特異的であるといえる。(他のACPsでも全くphosphopantetheinylateできないわけではない。)

FdmWとの酵素活性
FdmH ACP, with a K(m) of 5.8 microM and a k(cat)/K(m) of 8.1 microM(-1) x min(-1)
TcmM ACP with a K(m) of 1.0 x 10(2) microM and a k(cat) /K(m) of 0.6 microM(-1) x min(-1)

他のPPTasesとのalignment結果からN末37aa truncated FdmWを作成して活性測定し、PPTase活性にはこのN末部分が必要であることを確認している。
