配列解析のみ。

PKS starter unitであるbenzoic acidの生合成経路(Figure 3)で、3番目の反応として
cinnamoyl-CoA→3-hydroxy-3-phenylpropionyl-CoAを触媒すると考えられている。

EncI: Cinnamoyl-CoA hydratase

encH, encI, encJの破壊mutantは、benzoyl-CoA-primed enterocinの収量がwildより下がった。
encI mutantは、cinnamic acid and enterocin産生が20-25%まで低下するが、d5-benzoic acidの補充でenterocin産生はwildレベルまで回復する。
完全に抑制されないのは、一次代謝酵素が代替しているのかもしれない。

EncI cinnamoyl-CoA hydrataseはfatty acid enoyl-CoA hydratasesにAA配列類似性があり、Gly-116, Glu-119, and Glu-139は相同なfatty-acid-type hydratasesの触媒活性に必要な残基である。

BCS operonを E. coliで大量発現して、crotonase, beta-hydroxybutyryl-CoA dehydrogenase activityを確認。

BCS operonを C. acetobutylicumで発現すると、crotonase, beta-hydroxybutyryl-CoA dehydrogenase, butyryl-CoA dehydrogenase activityが上昇。
