Benastatin A and B生合成をコードする全遺伝子座のクローニング、シークエンス。
flanking regionsの削除とS. albus, S. lividansでの異種性発現によってclusterが同定されている。

BenF: methyl transferase

Benastatin生合成のlast stepはrings A and Cの間のmethylene bridgeのアルキル化。
feeding studiesによって2つのSAM由来methyl基がこのpositionに導入されると明らかになったが、ben clusterにmethyltransferaseはひとつのみ。
機能実験により、BenFが珍しいgeminal bis-methylationをできることが明らかになった。(unpublished observations)

benFありなしでben clusterを異種性発現、産物解析、相補確認。
benFがないとBenastatinやanthrone-like中間体は検出されず、quinone派生体とbis-nor-benastatin dimersを形成。

よってBenFのみでbismethylationを触媒することを確認。
このmethylationはoxygenationと競合する。
