aureolic acid type antitumor drug chromomycin A3の生合成gene clusterの同定論文。

CmmA: O-acyltransferase

配列解析から機能推定。CmmAは、macrolide aglyconeでも、macrolide aglycone付着済みのsugarsでも働くacyltransferasesに似ている。
よってdisaccharide鎖の1つめの糖D-olioseのC-4 OH基 and/or trisaccharide鎖の3つめの糖L-chromoseのC-4' OH基にacetyl基を移すと考えられている。

cmmA不活化mutantは、chromomycin A3に含まれる2つのacetyl基を両方持たないdideacetylated chromomycin A3派生体を蓄積。この化合物は、cmmA発現S.albusにおいて4E-monoacetylated中間体を介してchromomycin A3へと変換される。
このacetylationはchromomycin生合成で最終stepであり、産生生物の自己抵抗性のために重要。

4E-のEはsugar E。trisaccharideの3rd sugar L-chromoseのこと。
つまり先にsugar E(L-chromose)が、次にsugar A(D-oliose)が4-OH基でacetyl基を取り込む。
