cosK: Glycosyltransferase

aclacinomycin生合成で2-deoxy-L-fucoseの付加を担うAknKに似ていることから、glycosyltransferase CosKはcosmomycin D生合成で2つめのdeoxysugarである2-deoxy-L-fucoseの取り込みを担うと考えられた。

これを検証するために作製されたcosK不活化mutantは、cosmomycinsではない4つのanthracycline化合物を産生。
これらはすべてaglyconeに付着したL-rhodosamineを含むので、CosKは糖鎖の2つめのdeoxysugarの移動に関与すると考えられる。

AknKを異種性発現して精製、実験に使用。
dTDP-L-2-deoxyfucoseとrhodosaminyl aklavinoneから、AknKはL-2-deoxyfucosyl-L-rhodosaminyl aklavinoneを生成。
dTDP-L-daunosamineなどでも作用し、anthracycline骨格で2糖目を組み立てる。
活性は下がるが、rhodosaminyl aklavinoneにdTDP-L-2-deoxyfucoseをtandemに追加して、L-deoxyfucosyl-L-deoxyfucosyl-L-rhodosaminyl aklavinoneを産生することもできる。
