Elloramycin A産生に関連するcosmid clone 16F4からの遺伝子同定、配列解析とその遺伝子の異種性発現。

ElmG: tetracenomycin B2 oxygenase

elloramycinのaglycon 8-demethyltetracenomycin C (8-DMTC)は、tetracenomycin Cと8-OH基が違うだけ。よってElmGは、tetracenomycin(Tcm) B2 → 8-DMTCへの変換を担うと提唱されている。
この反応は、C-4, C-4a, C-12aでのtriple hydroxylation.

ElmGの基質として提唱されているTcmB2は入手不可能なので、ElmG酵素活性測定の基質にはTcmA2を使用。S.lividansで過剰発現、精製されたElmGは、TcmA2を同じ場所でtriple hydroxylationしたTcmCへと変換することをHPLC測定にて確認している。NADP or NADPHをelectron donorとして利用し、O2分子を必要とする。

8-DMTCがelloramycin aglyconeであることが確認されていることからTcmB2が直接triple hydroxylationされると判断し、基質名をTcmB2としている。
