Medermycinはその強力な抗癌活性に重大なピラン環において、actinorhodinと同じtrans立体配置(3S, 15R)を示す。
actinorhodin生合成において、ActVI-ORF1がC-3での立体化学を決定すると既に証明されている。
actVI-ORF1不全のある株をmed-ORF12で機能的に相補できた。

actinorhodinの早期段階生合成遺伝子とmed-ORF12の同時発現から、med-ORF12がピラン環の形成においてC-3でエナンチオ選択性還元を担うketoreductaseをコードすると実証。
