R1128 gene clusterがあると予測されるStreptomyces sp. R1128の40kb領域をS.lividans K4-114で異種性発現し、産物としてR1128Cと新規産物HU235を検出。
この領域をシークエンスして、生合成genesがすべて含まれていそうな17kb領域をサブクローニング、oligonucleotide walkingして正確な配列を得た。

zhuN: Acyl carrier protein (ACP)

zhu clusterにACP候補は2つあるが、配列解析からこちらがminimal PKSに含まれるACPであると推定している。

R1128 gene clusterにCPが2つあることに関する分子的ロジックを検証するため、apo-ZhuG or apo-ZhuNの存在下で、ZhuHが触媒するbeta-ketopentanoyl-ACP形成の抑制を定量化。

両apo-proteinsは同程度のinhibitorsであったので、これらはchain initiationの際に互換的に使用されうる。

minimal PKSのACPであるFrenN, ZhuN, DpsGの存在下では検出されない。
