streptomycinを産生するStreptomyces griseusのstrD and strEをプローブとして、mithramycinの糖部分の生合成に関する遺伝子(mtmD, mtmE)をmithramycin産生株S. argillaceusのcosAR7から分離。

MtmDは抗生物質生合成に関連したstreptomycetesのputative glucose-1-phosphate:TTP thymidylyl transferases(StrD, GraD, SnoD, DnrL)に似ている。

mtmD and mtmEをE.coliで一緒に発現し、その細胞抽出液を用いてdTDP-D-glucose synthase活性、dTDP-D-glucose-4,6-dehydratase活性を測定。IPTGでの遺伝子発現誘導で両酵素活性は3-4倍に増えた。

mtmDを置換して不活化したmutantは、糖を持たない4環のpremithramycinoneを蓄積した。よって、いくつか(またはすべて)のglycosylationsは4th ringの切断の前に起こると示唆される。
