streptomycinを産生するStreptomyces griseusのstrD and strEをプローブとして、mithramycinの糖部分の生合成に関する遺伝子(mtmD, mtmE)をmithramycin産生株S. argillaceusのcosAR7から分離。

MtmEは抗生物質経路のTDP-D-glucose 4,6-dehydratases(StrE, TylA2, GraE, Gdh)にとても似ている。

mtmD and mtmEをE.coliで一緒に発現し、その細胞抽出液を用いてdTDP-D-glucose synthase活性、dTDP-D-glucose-4,6-dehydratase活性を測定。IPTGでの遺伝子発現誘導で両酵素活性は3-4倍に増えた。
