使用されたS. rimosusの株はoxytetracycline(OTC)の商業的産生に使用される原栄養株M4018にすべて由来している。

OTCに感受性の自然発症S. rimosus mutantに耐性を与えるDNA断片から、OTC耐性決定因子otrAをクローニングしている。

OTC産生細菌S. rimosusにおけるOTC-resistance gene, otrAのcoding and regulatory regionsの配列を測定。配列解析から、GTPの結合と加水分解がタンパクの機能に重要であると考えられる。

otrAに特有な唯一のpromoter otrAp1は両方向へのpromoter活性があり、OtrAがあると反対向きの転写発現は抑えられる。

この論文より前に、OTC gene clusterを挟むように2つのOTC-resistance genes (otrA, otrB)が同定されており、これらが独立してS. lividans and S. albusでOTC耐性を与えることがわかっている。OtrAは非共有結合性修飾によりホストのリボソームに抵抗性を与えることも報告されている。
