benzoxazolinate生合成に関与するSgcD,SgcGの機能解析

SgcGは、相同性を示すIsf and WrbA proteinsと同様にN末にはflavodoxin-like domainを保存し、iron coordinationに関与する4つのcysも保存していた。

SgcGをcloning,大腸菌にて発現・精製し活性を調べている。
HPLC解析は、sgcGの濃度依存的に3-enolpyruvoylanthranilate(OPA)の形成を確認した。また、唯一の補基質としてFMNが同定された。
反応速度論的解析にてこの結果をサポートしている。
