C-1027のbeta-amino acid moiety生合成の報告

sgcC欠損株は22-Deshydroxy-C-1027を産生することが以前の実験[PMID:12183628]で示されていると明記あり。
SgcC2のFT-MSの結果と、SgcC1のアミノ酸依存性のATP-[32P]PPi exchange dataは、SgcC1が(S)-beta-tyrosine特異的なadenylation enzymeで、SgcCとSgcC3はbeta-aminoacyl-S-SgcC2を基質として利用することを明らかにした。

SgcCの機能解析

SgcCはC-1027へ取り込まれる前に(S)-3-chloro-5-hydroxy-beta-tyrosine moietyの生合成の最終段階として(S)-3-chloro-beta-tyrosyl-S-SgcC2のhydroxylationを触媒する酵素であると示した。
SgcCはflavin reductase component(sgcE6)からreduced flavin(FADH2)を得てtwo-component systemとして機能し、芳香環の5位でhydroxyl基の挿入を行うと示した。
carrier-protein-tethered aromatic substrate上で機能するtwo-component, FAD-dependent monooxygenaseとして初めて機能解析された例である。
