warhead cassette(UNBL-UNBV-UNBU-PKSE-TEBC)の報告

warhead cassette(UNBL-UNBV-UNBU-PKSE-TEBC)はactinomyceteにおいて全てのenediyne生合成遺伝子座で保存されているが、機能は未知のままである。enediyne warheadのformation, stabilization, or transport に関与しているのではないかと推測している。C-1027ではTEBC(thioesterase)のみが離れて存在する。

UNBVの構造解析は、N末端シグナル配列と共にsecreted proteinを持っていた。

dynE8, orf8, dynU15, dynU14 and orf23の欠損株を作製し、培養液中の産物をHPLCにて検出。orf8はdynemicin Aが検出できたが、dynE8, dynU15, dynU14,orf23の欠損株はdynemicin Aが検出できなかった。ただし、orf23の欠損株はdynemicin Aの新規化合物が検出された。
よって、dynE8, dynU15, dynU14はdynemicin Aに必須であることを示した。

dynU15, U14, T3, E8, E7は、minimal enediyne cassetteとして知られている遺伝子。
DynU15, U14, T3はconserved unknown proteins。
