CalG1, CalG4の機能・構造解析

CalG4を大腸菌にて発現、精製。
dTDP存在下でCalG4はcalicheamicin誘導体からaminopentose sugar moietyの切り離しを触媒した。対照としたdTDPなし or CalG4なしではstarting materialのみ検出した。よって、CalG4がcaricheamicin生合成に関与するaminopentosyltransferaseであることが示され、aminopentose sugar moietyが以前予測されていたUDP-sugar pathwayではなく、dTDP-sugar pathwayを経由したaminopentose生合成経路であることを示した。
