For vial culture, dissolve all ingredients except cysteine-HCl, flush N2/CO2, dispense 20 ml/50-ml vials, fix with a stopper with flushing N2/CO2 and autoclave. After autoclaved, the gas phase is replaced with H2/CO2 (80/20, 150 kPa), and add 0.1 ml of autoclaved cysteine-HCl solution (50 g/l) per each vial separately. Inoculum size 5-10%. |
