Composition
KH2PO4 0.136 g   NH4Cl 0.54 g   MgCl2·6H2O 0.2 g   CaCl2·2H2O 0.147 g   NaHCO3 2.5 g   Bacto Yeast Extract (Difco) 0.2 g   Vitamin solution* 10 ml   Trace element solution** 10 ml   Resazurin 1 mg   Cysteine-HCl 0.5 g   Na2S·9H2O 0.5 g   Distilled water 1 L     Mix ingredients except vitamin solution, cysteine-HCl and Na2S·9H2O, and autoclave under a H2/CO2 atmosphere (80/20). Concentrated cysteine-HCl and sodium sulfide solutions are sterilized separately under a N2 atmosphere in tightly closed vessels. Prior to inoculation, add the filter-sterile vitamin solution, Cysteine-HCl and Na2S·9H2O. Pressurize the inoculated vessels to 150 kPa with H2/CO2 (80/20).                   *Vitamin solution       Biotin 2 mg       Folic acid 2 mg       Pyridoxine-HCl 10 mg       Thiamine-HCl 5 mg       Riboflavin 5 mg       Nicotinic acid 5 mg       Ca-pantothenate 5 mg       p-Aminobenzoic acid 1 mg       Vitamin B12 0.01 mg       Distilled water 1 L                   **Trace elements solution       2Na-EDTA 0.64 g       MgSO4·7H2O 6.2 g       MnSO4·4H2O 0.55 g       NaCl 1 g       FeSO4·7H2O 0.1 g       CoCl2·6H2O 0.17 g       CaCl2·2H2O 0.13 g       ZnSO4·7H2O 0.18 g       CuSO4 0.05 g       KAl(SO4)2·12H2O 0.018 g       H3BO3 0.01 g       Na2MoO4·2H2O 0.011 g       NiCl2·6H2O 0.025 g     Dissolve the 2Na-EDTA in 500 ml of distilled water, and after adding the salts, make up to 1 L.
