Borrelidinはtrans(module 2)とcis(module 3)と両方の二重結合を持つ。

ACP結合中間体の代理基質としてSNAC thioesterを用いて、BorDH2とBorDH3のin vitroでの変換能力をみている。

BorDH2の代理基質として3-hydroxybutanoic acid SNAC thioesterを使ったとき、BorDH2は(3R)-だけで触媒してtrans産物を導いた。BorDH3ではどちらもほとんど変換できなかった。

BorDH3の代理基質として3-hydroxy-2-methylpentanoic acid SNAC thioesterを使ったとき、BorDH2とBorDH3は両方とも(2R3R)-をtrans産物に変換した。

この結果を受け、cis二重結合ができるメカニズムについていくつかの可能性を挙げている。
