CrpA: type I PKS

cryptophycin1のaglycon形成には2つのPKSと2つのNRPSが関与。
PKS(crpA-B)はunitA部分(sigma-hydroxy-phenyloctenoic acid)の形成を担う。
 
crpAはmodular type I PKSのloading domain, module 1をコードする。
loading domainはadenylation domain + ACP domain

ラベルしたphenylalanineの取り込み実験(Nostoc sp. GSV 224で行われている)の結果からphenylacetyl-CoAがstarter unit候補とされたが、ラベルしたphenylacetic acid と対応するN-acetylcysteamine thioesterを与えた実験で取り込みは示されなかった。AA特異性コードが一致しているMcyGでの実験報告に基づき、trans-cinnamic acidがstarter unitとして機能する可能性があると述べられている。

取り込み実験はacetate, methionineでも行われているが、Nostoc sp. GSV 224を用いている。
