4つのputative regulatory genes(fscRI, fscRII, fscRIII, and fscRIV)の推定産物はすべてtranscriptional regulatorsのLuxR familyに属しており、FR-008生合成の調節に関与しているかもしれない。
そのようなtranscriptional regulatorsは大きなPKS genesの非常に長いmRNAsの安定性を維持するために働くかもしれない。

fscRII、fscRIIIと部分的なfscRIを含む部分を置換したmutantでは、いかなるFR-008-related compoundsも産生しなかったことから、putative regulatory activitiesが裏付けられた。

regulatory genes候補の不活化mutantにおいて、nystatin gene cluster内にあるputative promoter regionsの調節下に置いたxylE reporter gene活性を測定。不活化mutantのCross-complementation experiments。

nysRIVはnysRI, RII, RIII のmutantsを相補できるのだが、逆にそれらのregulatorsはnysRIV mutantを相補できないので、nysRIVは最も直接的にnystatin生合成遺伝子の発現を調節しているのだろう。
