Rubradirin生合成clusterの報告。

配列比較からのProposed function
RubN8(424aa): Unknown

RubN1-N8はdTDP-D-rubranitroseの合成に推定的に関与していることが配列解析で明らかになった。

推定的にRubN8をamine oxidaseに割り当て。RubN8はrubradirin aglyconでのD-rubraminoseのglycosylationの後に、dTDP-D-rubranitrose生合成の最終ステップで働くことが考えられる。

その後のニトロソ基→ニトロ基への酸化は空気によって容易に起こることがわかっている。

dTDP-evernosamine → dTDP-evernitrososeへのflavin-dependent double-oxidationをすることを、反応産物をelectrospray HPLC/MS、Tandem MS fragmentationで解析することによって確認している。

amine→nitroso官能基への4電子のflavin-mediated oxidationは2stepsで起こる。
1)amino sugar (-NH2)→ hydroxylamino sugar (-NH-OH)
2)hydroxylamino sugar (-NH-OH) → nitroso sugar (-NO)
この両方を行うのがRubN8.
