resistomycin生合成gene cluster全体を異種性発現により同定。

RemH: Methyltransferase

PKS後の修飾として、bis-nor-resistomycinはgeminal bis-methylationを受ける。既報の13C-labeling実験でresistomycinの両方のgem methyl基がS-adenosyl methionine(SAM)に由来することがわかっている。remG, remHにはprobable SAM-binding methylTFに特徴的なmotifが保存されているので、この反応に関与することが示唆される。

delta-remGHはresistomycin非産生。remGH、remGで相補可能だがremHでは相補不可。
delta-remGもresistomycin非産生。remGで相補可能。
delta-remHはresistomycin産生可能。

remGを欠いたmutantでは不安定な化合物2を検出。
この化合物2がgeminal bis-methylationを欠いている構造であることを確認。

よってRemGのみがresistomycinに2つのmethyl基を導入するのに必要とされる。
