Daunorubicin (DNR) 生合成PKSの上流領域にある6ORFsの同定。

orf1: Ketoreductase
similar to Streptomyces antibioticus Orf4 (30%)

Orf1はketoreductase候補だが、NADPH存在下でaklaviketone → aklavinoneの還元を2.9%しかできない。promoter下にorf1またはdauBを置いて過剰発現したplasmidでは、dauE(aklaviketone reductase) mutant strainを相補できない。

DNR生合成で知られたketoreductase-like functionで唯一残っているのはC-13 keto基の還元であるが、orf1がこの機能をコードするかどうかは今のところ分かっていない。→続報も出ていない

dnrU破壊株の代謝産物は、wildと比べて、doxorubicin (DXR)が約3.4倍、RHOが約5.8倍に増える。
dnrX mutantではDXR産生が3倍に、RHOは検出されず。

dnrX dnrU double mutantはDXR産生で加算的な効果がある。
dnrX dnrU dnrH triple mutantは、dnrX dnrU double mutantより3.4-3.8倍多いDXRを産生。13-Deoxy-DNRも13-dihydro-DNRも産生されない。

これらのデータから完全な説明はできないが、DnrUはdaunorubicin (DNR) or DXR → anthracycline系代謝のその他の産物への経路が考えられている。
