dTDP-deoxyglucose epimerase geneをコードすると思われるgerFをE.coliで発現、精製、機能確認。
精製された酵素はdTDP-4-keto-6-deoxyglucoseからmaltolを産生し、よって発現されたタンパクがdTDP-4-keto-6-deoxyglucoseのC-3 and C-5 or C-3の異性化を触媒するdTDP-deoxyglucose epimeraseであることが確認された。

gerK1をクローニング、E.coliで過剰発現、機能確認。
GerK1はC-3でaxialなOH基を持つときのみ、dTDP-4-keto-6-deoxy-D-alloseの4-keto基で特異的な還元効果を示す。
GerFの存在下で、GerK1はdTDP-4-keto-6-deoxyglucose → dTDP-6-deoxy-beta-D-alloseへの変換を触媒した。
このdTDP-6-deoxy-D-alloseの分離成功は、GerFがmycinose生合成経路において3,5-epimeraseではなく、3-epimeraseとして機能することを確認するのに十分な証拠を供給する。

S.lividansでS.avermitilis avrBCDEFGHIを発現させて外因性aglyconeと反応させて産物解析。
delta-avrFな発現株では、C-13にD-olivoseが1つbeta-glycoside結合したavermectin A1a3を産生する。
avermectin A1a3を産生できるのはdelta-avrFのみ。
C-3での立体化学は最終的に3-O-methyltransferaseによって決定されるので、AvrF proteinは5-epimeraseであって3,5-epimeraseではない。
