[PMID:23692164](2013) abstract
6-deoxyerythronolide B synthase由来のTE domainを含むrecombinant LipPks1をE.coliで産生させ、その基質特異性をin vitroで検証。

isobutyryl-CoAを含むいくつかのacyl-CoAsがstarter基質として受け入れられたが、acetyl-CoAでの産物は観察されなかった。

[PMID:23692164]において、LipPks1はisobutyryl-CoAを含むいくつかの異なるacyl-CoAsをstarter基質として受け入れることができ、その中でも2-methylbutyryl-CoAのkcat/Km値はisobutyryl-CoAの値に匹敵することが報告された。

そこで2-methylbutyryl-CoAのよく知られた前駆体であるisoleucineの存在下でS. aureofaciens Tu117を育てたところ、新規alpha-lipomycin類似体 21-methyl-alpha-lipomycinを分離することができた。この類似体は、alpha-lipomycinと変わらない抗菌活性を示した。

このように、種々の同定可能な薬剤候補を増やして薬剤発見を促進するためには、PKSs loading didomainのin vitro kinetic parametersを測定することが重要である。
