rimJ; crotonyl-CoA reductase

rimJ自体の機能解析は行われていないが、相同性よりcrotonyl-CoA reductaseと予想している。
rimJ不活化変異株では、rimocidin産物の割合が下がり、一方でCE-108産物の割合が上昇していることが示されている。

rimJ破壊株で産生される誘導体についての解析。それらのうちのひとつCE-108Dは、PKS伸長ステップ13でethylmalonyl-CoAの代わりにmethylmalonyl-CoAが取り込まれた構造だった。CE-108Dはcluster外にあるpolyene carboxamide synthase PcsAにより、改善された薬理学的特性を持つCE-108Eへと変換される。

Streptomyces collinusよりcrotonyl-CoA reductaseを精製し、諸性質を調べている。(ホモダイマーであること、crotonyl-CoA, short-chain-length (C4), enoyl-CoA compoundに対して高い基質特異性を示す一方で、shorter-chain-length (C3), enoyl-CoA thioester, acryloyl-CoAに対しては活性を示さない。)
また、crotonyl-CoA reductase遺伝子をクローニングしてE. coliで発現させて精製し、活性を確認している。
