orf4はactinorhodin polyketide chainのassembleにおいてbeta-keto groupを還元するbeta-ketoacyl--ACP reductase と弱い相同性を示す。活性残基やNAD+ binding siteはこのORFでも保存している。

barS1の機能解析の報告

本論文にてORF名を orf4->barS1 変更している。
recombinant酵素精製し、(±)-6-dehydro-VB-Aに対しての酵素活性を見ている。また、barS1欠損株は、virginiae butanolide(VB),virginiamycin生産能が欠けていた。VBの外因的添加によりvirginiamycin生産能が回復したことから、virginiamycinの生産にVBの生産が影響すると推測した。

酵素学的解析からBarS1は、VB biosynthesisの最終ステップ6-dehydro-VB-A->VB-Aへの変換に関与する唯一の6-dehydro-VB-A reductaseであると示した。
