S. lividansで、AT domainsのひとつがmethoxymalonateに特異的なATで置換されているerythromycin PKSのカセットとasm13-17を同時発現すると、予測された位置にmethoxymalonateが取り込まれた6-deoxyerythronolide Bの派生体が産生された。よって、asm13-17はmethoxymalonyl-ACP生合成に十分である。

また、methyltransferaseをコードするasm17を削除すると、そのhydroxy類似体ではなく6-deoxyerythronolide Bが得られたので、伸長unitのmethylationはその取り込みのために必要である。

asm13-17はオペロンを形成。asm13 と asm14はoverlapsがあるので転写共役するかもしれない。
稀なpolyketide鎖伸長unit methoxymalonayl-ACPの形成に関連すると提唱されている。
