ΔrevJではReveromycinの生産物が25%以下になり、相補株では生産能が回復した。
RevGの反応産物から、立体構造の異なるスピロアセタール化合物(A3a(18a)とA3b(18b))が非酵素的に生成されるが、Streotimyces sp. SN-593から単離されるReveromycinはA3a(18a)由来のみである。
RevGの反応mixtureに精製RevJを加えたところ、RevJが反応系に存在するときのみスピロアセタール環化反応が効率よく進行し、さらに反応産物の立体構造はすべてA3a(18a)だった。
安定な非環式の前駆体から、スピロアセタール立体構造を構築するkey酵素がdehydroxy ketone synthase(RevG)及びspiroacetal synthase(RevJ)である。
