S.albusに外因性mithramycinへの抵抗性を与えるS. argillaceusの最小DNA領域(3.9kb)がシークエンス、配列解析され、不完全なORF(mtrX)と完全ORFs 3つ(mtrY, A, B)が予測された。

mtrYの推定産物は、DBに明らかな類似性を示すものなし。

mtrX と mtrYの各frameshift mutantは、外因性mithramycinに対する抵抗性に影響がない。しかし、内因性に合成されたmithramycinに対する防御と分泌に関与するかもしれないという可能性は残っている。

mtrYの遺伝子置換による不活化でmithramycin生合成は50%減。
mtrYを過剰発現するとmithramycin生合成は約47%増。

よって、mtrYはたぶんpositive regulatory elementをコードし、その発現は普通レベルのmithramycin産生に重要であると強く示唆される。

N末近くにはputative helix-turn-helix motif (HTH)あり。
