oxyQ (otcX2): Aminotransferase

アミノ酸代謝に関連するaspartate/tyrosine/aromatic aminotransferasesに相同であることから、pyridoxal 5'-phosphateをcofactorとして4-keto-ATCのC-4にアミノ基転移する酵素のようだ。

完全に機能的なring Aを含む最初の中間体anhydrotetracycline (ATC)の生合成に必要な最小酵素セットの同定と再構築。

in vivo and in vitro approachesを組み合わせて、6-methylpretetramid → ATC への変換に必要なのはOxyL, OxyQ, OxyTであることを確認した。

OxyLはNADPH-dependent dioxygenaseであり、6-methylpretetramid に2つの酸素原子を導入し、不安定な中間体4-keto-ATC をもたらす。
aminotransferase OxyQは4-keto-ATC の還元的アミノ化を触媒し、4-amino-ATC をもたらす。
さらに、N,N-dimethyltransferase OxyTはSAM-dependent様式で、4-amino-ATC → ATC の形成を触媒する。

CH999/pWJ135(OxyABCDJKNF + OxyL)で、4-keto-ATCの分解産物を確認。
CH999/pWJ209(OxyABCDJKNF + OxyL + OxyQ)では、4-amino ATCを蓄積。
よってOxyQはreductive transaminaseであると確認された。
