mdpC2: NRPS peptidyl carrier protein

Eight genes(mdpC to mdpC8 excluding mdpC5)は、L-alpha-tyrosineからの(S)-3-(2-chloro-3-hydroxy-4-methoxyphenyl)-3-hydroxypropionic acid moietyの生合成に関与するとしている。

L-alpha-tyrosineがmdpC4により触媒され(S)-beta-tyrosineになった後、活性化されfree-standing MdpC2 PCPが(S)-beta-tyrosineに付着すると推測。

sgcC1とsgcC2は大腸菌で発現・精製、過剰発現させた。
apo-sgcC2をCoA存在下でphosphopanteinyl transferaseと反応させた時のFT-MS値は、active site Serineがphosphopantheinylationされた時の計算値と一致した。また、holo-sgcC2にATP存在下で7(SgcC1反応産物)、3(tyrosineとSgcCの反応産物)とSgcC4のmixtureをそれぞれ反応させると、(S)-beta-tyrosinyl-S-SgcC2中間体のmassと一致したことを見ている。
