mdpC1: NRPS adenylation enzyme

Eight genes (mdpC to mdpC8 excluding mdpC5)は、L-alpha-tyrosineからの(S)-3-(2-chloro-3-hydroxy-4-methoxyphenyl)-3-hydroxypropionic acid moietyの生合成に関与するとしている。

free-standing peptidyl carrier protein(PCP)(MdpC2)への(S)-beta-tyrosineのloadingは、C-1027のSgcC1と相同性を示すMdpC1によって行われると推測している。

amino acid-dependent ATP-PPi exchange assayは、MdpC1がL-alpha-tyrosineを認識出来ず、MdpC1を(S)-beta-tyrosine とincubateした時に活性が確認された。MdpC1はSgcC1と同様に他のbeta-amino acid基質とも反応した。
MdpC1は(S)-beta-tyrosine-activating adenylation enzymeであると結論付けている。

sgcC1とsgcC2は大腸菌で発現・精製、過剰発現させた
アミノ酸依存性のATP-[32P]PPi exchange dataからsgcC1は(S)-beta-tyrosineに特異的なadenylation enzymeであることを示した。

SgcC1の機能解析

SgcC1はL-alpha-tyrosineを活性化するA domainと相同性を示した。nonribosomal codesのアライメント比較から、SgcC1のAsp235とLys517はalpha-amino and the carboxylate groupsと相互作用すると推測された。また、SgcC1の基質特異性は、alpha-amino acidsではなくbeta-phenyl-alanine or beta-tyrosine analogsのみを認識、特に(S)-beta-tyrosineがSgcC1の基質としてもっとも好ましかった。
