C-1027のbeta-amino acid moiety生合成の報告

SgcC2のFT-MSの結果と、SgcC1のアミノ酸依存性のATP-[32P]PPi exchange dataは、SgcC1が(S)-beta-tyrosine特異的なadenylation enzymeで、SgcCとSgcC3はbeta-aminoacyl-S-SgcC2を基質として利用すると示した。
また、sgcC3,sgcCの各欠損株の産物より、SgcC3はSgcCよりも前に働くと推定している。

SgcC3の機能解析

SgcC3 は、L-alpha-tyrosineから合成される(S)-3-chloro-4,5-dihydroxy-β-phenylalanine moietyの生合成において3番目のenzymatic transformationを触媒する事を明らかにした。
SgcC3はflavin reductase component(sgcE6)からreduced flavin(FADH2)を得てtwo-component systemとして機能し、SgcC2につながれた基質のchlorinationとbrominationを行うことを示した。
