CalG1, CalG4の機能・構造解析

aglyconeにdTDP-beta-L-rhamnoseとCalG1を反応させると、新規化合物が産生する。aglyconeにdTDP-beta-L-rhamnoseのみを反応させると化合物は検出されない。したがって、この反応にはCalG1が必須であることを示した。

また、dTDP-beta-L-rhamnoseをdTDP-alpha-L-rhamnoseに替えて反応させると化合物は検出されないことから、CalG1は立体特異的な転化型のglycosyltransferaseだと判明した。

CalG1のNDP-sugar特異性を22種のdTDP sugarを使って調査。この結果は、CalG1がcalicheamicin rahmnosyltransferaseであり、dTDP-D- and dTDP-L-sugar donorに対してフレキシブルであることを示した。

さらに、CalG1はdTDP存在下でnative calicheamicin 3'-O-methylrhamnosyl unitを効率的に切りはなすことも示した。
