CalG3, CalG2の機能構造解析

CalG2を大腸菌にて発現、精製。
22種のdTDP-sugar基質を用いて化合物9とcalG2をincubateしたところ、dTDP-4,6-dideoxya-D-glucose(化合物12)でのみ新規産物が得られた。CalG2 acceptor nucleophileがマスクされている化合物8とdTDP-4,6-dideoxy-alpha-D-glucoseはCalG2では触媒できなかったことから、CalG2は化合物9に糖を付加することが出来ると推測。したがって、CalG2は4-deoxy-4-thio-alpha-D-digitoxosyltransferaseであるとしている。
