CalO1の機能解析

cofactor(S-adenosylhomocysteine)に結合したCalO1を構造解析(2.4 Å resolution)。CalO1とCalO6の立体構造比較を行っている。CalO1とCalO6基質は似たような分子構造を共有していると思われ、基質結合領域のいくつかの残基の違いが基質再配向に影響を与え、その結果2つの酵素間で位置特異性の違いを生むと推定される。

CalO6はin vitroでN-acetyl cysteaminyl orsellinic acid(SNAc-orsellinic acid)のC2 OHの効率的なメチル化を証明した。(S. Singh & J. S. Thorson, unpublished work)。

この調査から、C2-O-methylationはC3-hydroxylation/methylation と C5-iodinationより前に起こり、CoAまたはACPが結合した基質を必要とすることが示唆された。
