Putative VanY-type carboxypeptidase

vancomycin様グリコペプチドであるbalhimycinの生合成gene clusterにあるaccessory resistance gene orthologs vanY, vnlR, vnlSの役割を調査。

VanYを過剰発現したE.coliの細胞抽出液は、diacetyl-Lys-D-Ala-D-Alaに対してかなりのDD-carboxypeptidase活性を示すが、diacetyl-Lys-D-Ala-D-Lacでは検出されなかった。よってVanYはDD-carboxypeptidase活性を持ちpeptidoglycan前駆体から末端のD-Alaを切断するが、DD-carboxyesterase活性を持たない。

vanHAX genesを誘導できるStreptomyces coelicolorでのvanYの異種性発現は、balhimycin耐性を拡大した。regulatory two-component system VanRSがないためvanHAX genesが誘導されないglycopeptide感受性mutant S.coelicolor J3201においてvanYを異種性発現しても、balhimycin耐性は与えられなかった。よってS.coelicolorにおいて、vanY単独では耐性を与えることはできず、機能的なvanHAX genesを必要とする。
