ORF28はputative hydroxylaseをコードする。ORF28のmutantはtyrosine部分でbeta-OH基を欠いているA40926のdesoxyderivativeを産生したので、ORF28がBht残基の形成に実際に関与することが実証された。また、このmutantに遊離Bhtを与えてもA40926産生は回復しないため、NRPS module 6の基質がBhtではなく遊離Tyrである可能性が示唆されるが、chlorinated Bhtである可能性も除外はできていない。
