A40926を産生するNonomuraea sp. ATCC 39727におけるグリコペプチド耐性の調査。
Nonomuraea sp. ATCC 39727に存在するpeptidoglycan前駆体は、pentapeptideの末端D-Ala残基が除去されたtetrapeptideが優勢であり、これはdbv gene clusterにあるvanY(ORF7)のD,D-carboxypeptidase活性に基づくとされる。
vanY-null mutantはグリコペプチド耐性レベルが下がる。
Streptomyces venezuelaeにおけるvanYの異種性発現は、グリコペプチド耐性レベルを高める。

E. coliで発現して得たVanY(n)を用いて活性確認。D,D-carboxypeptidase活性と D,D-dipeptidase活性を示すが、D-Ala-D-Lacで終結したペプチドでの D,D-carboxyesterase能力を欠く。VanY(n)の活性は非競合様式でβ-ラクタムにより抑制される。
